Tetranitro-blue Tetrazolium Reduction in Proteus Vulgaris
نویسندگان
چکیده
In Bacillus subtilis the reduction of potassium tellurite and of tetranitro-blue tetrazolium (TNBT) give comparable results: reduced products appear deposited in particular membranous organelles. For this reason these specialized organelles are considered to be chondrioids (1-3). In the Gram-negative Proteus vulgaris similar membranous structures were never observed by us (4, 5). Therefore it is a challenging problem to investigate the location of the respiratory chain in this organism. With potassium tellurite applied to Proteus (2, 4, 5) in a manner similar to that for Bacillus subtilis (1, 2, 5), we observe deposition of reduced product in a rather restricted cytoplasmic area contiguous to the plasma membrane. In view of modern ideas that enzymes belonging to the oxidative phosphorylation process would be located in elementary particles forming part of the cristae mitochondriales, it was considered essential to determine upon what cell structures in this organism the products of tetranitro-blue tetrazolium reduction are deposited.

Tetranitro-blue Tetrazolium Reduction in Bacillus Subtilis
In order to decide whether in the bacterial cell the enzymes of the respiratory chain are located in "particulate elements" (1) or in the plasma membrane (2) we investigated the reduction of potassium tellurite in Gram-positive and Gram-negative bacteria. For the first time in 1962 (3), and in more detail in 1964 (4), we reported that with Bacillus subtilis we had found that reduction products ...
متن کاملThe Ciprofloxacin Impact on Biofilm Formation by Proteus Mirabilis and P. Vulgaris Strains
BACKGROUND Proteus spp. bacilli belong to opportunistic human pathogens, which are primarily responsible for urinary tract and wound infections. An important virulence factor is their ability to form biofilms that greatly reduce the effectiveness of antibiotics in the site of infection. OBJECTIVES The aim of this study was to determine the value of the minimum concentration of ciprofloxacin t...
متن کاملThe Demonstration of the Succinic Dehydrogenase System in Bacillus Subtilis Using Tetranitro-blue Tetrazolium Combined with Techniques of Electron Microscopy
Activity of the succinic dehydrogenase system was studied in Bacillus subtilis utilizing combined techniques of cytochemistry and electron microscopy. Organisms were incubated in a medium containing tetranitro-blue tetrazolium (TNBT) which served as an electron acceptor. Enzymatic activity, as evidenced by deposition of TNBT-formazan, was found on membranous organelles associated with the cytop...
متن کاملThe Use of Tetranitro-blue Tetrazolium for the Cytochemical Localization of Succinic Dehydrogenase
Succinic dehydrogenase (SDH) was localized in the mitochondria of Sarcoma 37 ascites tumor cells by the use of tetranitro-BT (TNBT) and nitro-BT (NBT) in smear preparations. Results with each tetrazolium salt as electron acceptor were evaluated with respect to: (a) size and shape of the formazan precipitate relative to standard mitochondrial morphology; (b) crystallization phenomena of reduced ...
